The Notch 1 receptor is cleaved constitutively by a furin like convertase . ^^^ We report here that constitutive processing of murine Notch 1 involves a furin like convertase . ^^^ Mutation of these four amino acids ( and of two secondary dibasic furin sites located nearby ) completely abolished processing of the Notch 1 receptor . . ^^^

Ligand induced signaling in the absence of furin processing of Notch 1 . ^^^ The primary 300 kDa translation product of the Notch 1 gene ( p 300 ) is cleaved by a furin like convertase to generate a heterodimeric , cell surface receptor composed of 180 ( p 180 ) and 120 ( p 120 ) kDa polypeptides . ^^^ Consistent with previous reports , we found that disruption of furin processing of Notch 1 , either by coexpression of a furin inhibitor or by mutation of furin target sequences within Notch 1 itself , perturbed ligand dependent signaling through the well characterized mediator of Notch signal transduction , CSL ( CBF 1 , Su ( H ) , and LAG 1 ) . ^^^

Using in vivo [ ( 32 ) P ] orthophosphate labeling of cells expressing transfected Notch 1 , we observed that the furin cleaved Notch 1 ( TMIC ) and the soluble intracellular forms ( NICD ) , but not the full length molecule were phosphorylated . ^^^

The NOTCH 1 receptor is cleaved within its extracellular domain by furin during its maturation , yielding two subunits that are held together noncovalently by a juxtamembrane heterodimerization ( HD ) domain . ^^^
