The present study demonstrates that erythropoietin ( Epo ) and IL 3 induce tyrosine phosphorylation of the SH2 / SH3 containing adapter protein CrkL and its transient association with tyrosine phosphorylated SHP 2 , Shc , and Cbl in a murine IL 3 dependent cell line , 32D , expressing the Epo receptor ( EpoR ) . ^^^ Studies using cells expressing mutant EpoRs showed that the Epo induced tyrosine phosphorylation of CrkL is dependent on the membrane proximal EpoR cytoplasmic region involved in the activation of Jak 2 as well as the C terminal 145 amino acid region which is required for tyrosine phosphorylation of SHP 2 and Shc . ^^^ It was further revealed that CrkL is recruited to the tyrosine phosphorylated EpoR , most likely through its interaction with tyrosine phosphorylated Shc and SHP 2 . ^^^

Here , we demonstrate that Epo induces binding of CrkL to the tyrosine phosphorylated EpoR and SHIP 1 in 32D / EpoR Wt cells overexpressing CrkL . ^^^ In vitro binding studies showed that the CrkL SH 2 domain directly mediates the EpoR binding , which was specifically inhibited by a synthetic phosphopeptide corresponding to the amino acid sequences at Tyr ( 460 ) in the cytoplasmic domain of EpoR . ^^^ Together , the present study suggests that , upon Epo stimulation , CrkL is recruited to the EpoR through interaction between the CrkL SH 2 domain and phosphorylated Tyr ( 460 ) in the EpoR cytoplasmic domain and undergoes tyrosine phosphorylation by receptor associated Lyn to activate the downstream signaling pathway leading to the activation of Erk and Elk 1 . . ^^^

We demonstrate here that cross linking of alpha4beta1 by anti alpha 4 antibody induces tyrosine phosphorylation of Pyk 2 , Shc , and Cbl as well as binding of the adaptor protein CrkL with Cbl in a murine hematopoietic cell line , 32D / EpoR Wt . ^^^
