| Specific motifs recognized by the SH 2 domains of Csk , 3BP2 , fps / fes , GRB 2 , HCP , SHC , Syk , and Vav . ^^^ We report here the optimal recognition motifs for SH 2 domains from GRB 2 , Drk , Csk , Vav , fps / fes , SHC , Syk ( carboxy terminal SH 2 ) , 3BP2 , and HCP ( amino terminal SH 2 domain , also called PTP1C and SHPTP 1 ) . ^^^ As predicted , SH 2 domains from proteins that fall into group 1 on the basis of a Phe or Tyr at the beta D 5 position ( GRB 2 , 3BP2 , Csk , fps / fes , Syk C terminal SH 2 ) select phosphopeptides with the general motif phospho Tyr hydrophilic ( residue ) hydrophilic ( residue ) hydrophobic ( residue ) . ^^^ |

| Adaptor function for the Syk kinases interacting protein 3BP2 in IL 2 gene activation . ^^^ Using a yeast two hybrid screen to identify Syk kinases interacting proteins ( SKIPs ) , we isolated 3BP2 , an Abl SH 3 interacting protein of unknown function . 3BP2 was selectively expressed in hematopoietic / lymphoid tissues and bound via its SH 2 domain activated Syk family kinases in mammalian cells , including in antigen receptor stimulated T cells . ^^^ This activity was dependent on the SH 2 and pleckstrin homology domains of 3BP2 , and required functional Syk kinases , Ras , and calcineurin . ^^^ Thus , 3BP2 is an important adaptor that may couple activated Zap 70 / Syk to a LAT containing signaling complex involved in TCR mediated gene transcription . . ^^^ |

| Adaptor protein 3BP2 , a c Abl Src homology 3 ( SH 3 ) domain binding protein , is known to play a regulatory role in T cell receptor mediated transcriptional activation of nuclear factor of activated T cell and activator protein 1 by interacting with Syk / ZAP 70 protein tyrosine kinase . ^^^ By using the transient expression system in COS 7 cells , we have demonstrated that 3BP2 was predominantly phosphorylated on Tyr 174 , Tyr 183 , and Tyr 446 when it was coexpressed with Syk . ^^^ |
