We have demonstrated previously that c Cbl requires the presence of a functional ubiquitin interacting motif ( UIM ) in Eps 15 to mediate epidermal growth factor receptor ( EGFR ) endocytosis . ^^^ Both the ubiquitin ligase activity of c Cbl and the UIM of Eps 15 were necessary for plasma membrane recruitment of Eps 15 and entry of ligand bound EGFR into coated pits and vesicles containing Eps 15 . ^^^ This is consistent with a scenario in which ubiquitin moieties appended to activated EGFR complexes act as docking sites for Eps 15 and thereby recruit receptors into clathrin coated pits . ^^^ We find that c Cbl can guide ligand bound EGFR into the Eps 15 internalization route by two distinct mechanisms . ^^^ No strict requirement exists for either ubiquitin modified EGFR or the Cbl binding ubiquitination substrate CIN 85 as docking site for the UIM of Eps 15 . ^^^

We have identified a regulated interaction between parkin and Eps 15 , an adaptor protein that is involved in epidermal growth factor ( EGF ) receptor ( EGFR ) endocytosis and trafficking . ^^^ Treatment of cells with EGF stimulates parkin binding to both Eps 15 and the EGFR and promotes parkin mediated ubiquitination of Eps 15 . ^^^ We propose that by ubiquitinating Eps 15 , parkin interferes with the ability of the Eps 15 UIMs to bind ubiquitinated EGFR , thereby delaying EGFR internalization and degradation , and promoting PI ( 3 ) K Akt signalling . ^^^
