Convergence of peroxisome proliferator activated receptor gamma and Foxo 1 signaling pathways . ^^^ Foxo 1 antagonized PPARgamma activity and vice versa indicating that these transcription factors functionally interact in a reciprocal antagonistic manner . ^^^ One mechanism by which Foxo 1 antagonizes PPARgamma activity is through disruption of DNA binding as Foxo 1 inhibited the DNA binding activity of a PPARgamma / retinoid 10 receptor alpha heterodimeric complex . ^^^ The Caenorhabditis elegans nuclear hormone receptor , DAF 12 , interacted with the C . elegans forkhead factor , DAF 16 , paralleling the interaction between PPARgamma and Foxo 1 . daf 12 and daf 16 have been implicated in C . elegans insulin like signaling pathways , and PPARgamma and Foxo 1 likewise have been linked to mammalian insulin signaling pathways . ^^^ These results suggest a convergence of PPARgamma and Foxo 1 signaling that may play a role in insulin action and the insulinomimetic properties of PPARgamma ligands . ^^^ |

METHODS : Glucose metabolism , insulin sensitivity , and gene expression of key adipocyte genes , including adiponectin , interleukin 6 ( Il 6 ) , 11 beta hydroxysteroid dehydrogenase ( 11beta Hsd ) , peroxisome proliferator activated receptor gamma ( Ppar gamma ) , forkhead box O 1 ( Foxo 1 ) , glucose transporter 4 ( Glut 4 ) , CCAAT / enhancer binding protein ( C / ebp alpha ) , and fatty acid synthase ( Fasn ) were characterized in adipocytes from epididymal and subcutaneous fat depots of 28 week old male WOKW rats and Dark Agouti ( DA ) controls . ^^^ The severe insulin resistance predominantly in epididymal adipose tissue of WOKW rats is associated with a 10 fold decrease in adipocyte adiponectin gene expression , decreased Ppar gamma , but increased Foxo 1 gene expression compared to DA rats . ^^^ |

FOXO 1 represses peroxisome proliferator activated receptor gamma 1 and gamma 2 gene promoters in primary adipocytes . ^^^ FOXO 1 and peroxisome proliferator activated receptor gamma ( PPARgamma ) are crucial transcription factors that regulate glucose metabolism and insulin responsiveness in insulin target tissues . ^^^ We studied the effects of FOXO 1 on human PPARgamma gene expression in primary rat adipocytes and found that both genes are endogenously expressed . ^^^ FOXO 1 coexpression dose dependently repressed transcription from either the PPARgamma 1 or PPARgamma 2 promoter reporter by 65 % , whereas insulin ( 100 nm , 20 24 h ) either partially or completely reversed this effect . ^^^ Phosphorylation defective FOXO 1 mutants T24A , S256A , S319A , and T24A / S256A / S319A still repressed the PPARgamma 1 promoter and partially lost their effects on the PPARgamma 2 promoter in either basal or insulin stimulated cells . ^^^ |
