Flt 3 signaling involves tyrosyl phosphorylation of SHP 2 and SHIP and their association with Grb 2 and Shc in Baf3 / Flt3 cells . ^^^ Like the chimeric murine Flt 3 , human Flt 3 undergoes autophosphorylation , associates with Grb 2 , and leads to tyrosine phosphorylation of Shc on ligand binding . ^^^ These results demonstrate that SHP 2 and SHIP are important components in the human Flt 3 signaling pathway and suggest that SHP 2 and SHIP , by forming complexes with adapter proteins Grb 2 and Shc , may modulate MAP kinase activation , which may be necessary for the mitogenic signaling of Flt3 . . ^^^

SHC and SHIP phosphorylation and interaction in response to activation of the FLT 3 receptor . ^^^ Intracellular FLT 3 signaling involves tyrosine phosphorylation of several cytoplasmic proteins including SHC . ^^^ We have found that upon FLT 3 activation SHC phosphorylation occurs at tyrosine 239 / 240 and 313 . ^^^ Overexpression of SHC increases the level of SHIP phosphorylation on tyrosines in response to FLT 3 activation , suggesting that SHC availability is a limiting step for SHIP phosphorylation . ^^^ Interestingly , SHC overexpression in FLT 3 activatable Ba / F3 cells limits FLT 3 dependent cell growth and this effect requires tyrosine 313 . ^^^
