Fluorescence resonance energy transfer studies on the interaction between the lactate transporter MCT 1 and CD 147 provide information on the topology and stoichiometry of the complex in situ . ^^^ The monocarboxylate ( lactate ) transporters MCT 1 and MCT 4 require the membrane spanning glycoprotein CD 147 for their correct plasma membrane expression and function . ^^^ We have successfully expressed CD 147 and MCT 1 tagged on their C or N termini with either the cyan ( CFP ) or yellow ( YFP ) variants of green fluorescent protein . ^^^ FRET was observed when either the C or N terminus of MCT 1 ( intracellular ) is tagged with CFP or YFP and co expressed with CD 147 tagged with YFP or CFP on the C terminus ( intracellular ) but not the N terminus ( extracellular ) . ^^^ Because FRET only occurs between two fluorophores if they are less than 100 A apart and in a suitable orientation , our data provide important information on the topology of CD 147 and MCT 1 within the plasma membrane . ^^^

After the detection of strong discrete granular cytoplasmic immunoreactivity in ASPS for monocarboxylate transporter 1 ( MCT 1 ) in the course of a broad immunohistochemical characterization of an MCT 1 antibody , we studied the expression of MCT 1 and its interacting partner , CD 147 , in a panel of 10 ASPS cases using appropriate antibodies . ^^^ We find that the periodic acid Schiff diastase resistant cytoplasmic granules of ASPS are strongly immunoreactive for MCT 1 and CD 147 . ^^^ Specifically , intense cytoplasmic granular positivity for MCT 1 and CD 147 was found in 7 of 10 and 8 of 10 ASPSs , respectively . ^^^ Western blot analysis of several ASPS cases confirmed that the protein reactive with the MCT 1 antibody and that reactive with the CD 147 antibody both migrated at the size expected for MCT 1 and CD 147 , respectively . ^^^
