METHODS : We investigated the uptake transporters OATP 2 ( SLC21A6 ) , OATP 8 ( SLC21A8 ) , and NTCP ( SLC10A1 ) , the export pumps MRP 2 ( ABCC 2 ) , MRP 3 ( ABCC 3 ) , MRP 6 ( ABCC 6 ) , and P glycoproteins ( ABCB 1 , ABCB 4 , ABCB 11 ) , and radixin , in non icteric primary biliary cirrhosis ( PBC stages 1 3 ) and control human liver needle biopsies using immunofluorescence microscopy and semi quantitative RT PCR . ^^^ Areas of irregular MRP 2 immunostaining showed largely reduced radixin immunostaining , whereas normal hepatocytes had MRP 2 and radixin confined to the canalicular membrane . ^^^ Disrupted localization of radixin and MRP 2 supports the concept that radixin contributes to the canalicular localization of MRP 2 . ^^^

Specific knockdown of Mrp 2 ( approximately 50 % decrease in expression ) resulted in an approximately 45 % decrease in the biliary excretion index of carboxydichlorofluorescein ( CDF ) ( 9 . 3 % versus 16 . 5 % ) , but did not affect Mrp 3 or radixin expression . ^^^
