Purified baculovirus expressed HsCdc 7 HsDbf4 selectively phosphorylates the MCM 2 subunit of the minichromosome maintenance ( MCM ) protein complex isolated by immunoprecipitation with MCM 7 antibodies in vitro . ^^^ Two dimensional tryptic phosphopeptide mapping analysis of in vivo ( 32 ) P labeled MCM 2 from HeLa cells reveals that several major tryptic phosphopeptides of MCM 2 comigrate with those of MCM 2 phosphorylated by HsCdc 7 HsDbf4 in vitro , suggesting that MCM 2 is a physiological HsCdc 7 HsDbf4 substrate . ^^^ These results indicate that the HsCdc 7 HsDbf4 kinase is directly involved in regulating the initiation of DNA replication by targeting MCM 2 protein in mammalian cells . . ^^^

Cdc 7 , Cdk 2 , and ataxia telangiectasia and Rad 3 related kinases regulate S phase entry and S phase progression and are known to phosphorylate the Mcm 2 subunit . ^^^ In this work , by in vitro kinase reactions and mass spectrometry analysis of the products , we have mapped phosphorylation sites in the N terminus of Mcm 2 by Cdc 7 , Cdk 2 , Cdk 1 , and CK 2 . ^^^ We found that Cdc 7 phosphorylates Mcm 2 in at least three different sites , one of which corresponds to a site also reported to be phosphorylated by ataxia telangiectasia and Rad 3 related . ^^^ Importantly , although all the Cdc 7 dependent Mcm 2 phosphosites fluctuate during the cell cycle with kinetics similar to Cdc 7 kinase activity and Cdc 7 protein levels , phosphorylation of Mcm 2 in the putative cyclin dependent kinase ( Cdk ) consensus sites is constant during the cell cycle . ^^^ Furthermore , our analysis indicates that the majority of the Mcm 2 isoforms phosphorylated by Cdc 7 are not stably associated with chromatin . ^^^

In this study , we have mapped the sites in human Mcm 2 protein that are phosphorylated by Cdc 7 . ^^^ Alanine substitution experiments with Mcm 2 peptides showed that the phosphorylation of ( 5 ) S and ( 53 ) S by Cdc 7 required the presence of an acidic amino acid adjacent to a serine residue . ^^^ Furthermore , although Cdc 7 was unable to phosphorylate a Mcm 2 peptide ( spanning amino acids 19 30 and containing ( 26 ) S and ( 27 ) S ) , it phosphorylated ( 26 ) S efficiently when this peptide contained a chemically synthesized phospho ( 27 ) S modification . ^^^ Hence , additional Cdc 7 phosphorylation sites could be generated in Mcm 2 by its prior phosphorylation by a cyclin dependent kinase . ^^^
