Beta arrestin 2 functions as a G protein coupled receptor activated regulator of oncoprotein Mdm 2 . ^^^ Oncoprotein Mdm 2 is a master negative regulator of the tumor suppressor p 53 and has been recently shown to regulate the ubiquitination of beta arrestin 2 , an important adapter and scaffold in signaling of G protein coupled receptors ( GPCRs ) . ^^^ However , whether beta arrestin 2 has any effect on the function of Mdm 2 is still unclear . ^^^ Our current results demonstrated that the binding of Mdm 2 to beta arrestin 2 was significantly enhanced by stimulation of GPCRs . ^^^ Activation of GPCRs led to formation of a ternary complex of Mdm 2 , beta arrestin 2 , and GPCRs and thus recruited Mdm 2 to GPCRs at plasma membrane . ^^^

In this study , we found that although both beta arrestin 1 and beta arrestin 2 are able to interact with ubiquitin protein isopeptide ligase ( E 3 ) Mdm 2 , only expression of beta arrestin 2 leads to the relocalization of Mdm 2 from the nucleus to the cytoplasm . ^^^ In addition , our data showed that nucleocytoplasmic shuttling of beta arrestin 2 was required , via protein / protein interaction , for the cytoplasmic relocalization of Mdm 2 and JNK 3 , another well known beta arrestin 2 binding protein . ^^^ Our study thus suggests that both the nuclear export signal motif and the N domain of beta arrestins are critical for the regulation of their subcellular localization and that beta arrestin 2 may modulate the function of its binding partners such as Mdm 2 and JNK 3 by alteration of their subcellular distribution . . ^^^
