Given that synprint has previously been shown to bind syntaxin 1A and that this association elicits inhibition of N type calcium channels , we tested whether htt ( exon 1 ) affects the modulation of these channels . ^^^ Our data indicate that htt ( exon 1 ) enhances calcium influx by blocking syntaxin 1A inhibition of N type calcium channels and attributes a key role for htt N terminal fragments in the fine tuning of neurotransmission . . ^^^

One candidate SERT regulatory protein is the SNARE ( soluble N ethylmaleimide sensitive factor attachment protein receptor ) protein , syntaxin 1A ( Syn1A ) , which has recently been implicated in the regulation of ion channels as well as the SERT related gamma aminobutyric acid and glycine transporters . ^^^

In the present report , the interaction of syntaxin 1A with endogenous serotonin transporters ( SERT ) expressed in developing thalamocortical neurons is examined . ^^^ Incubation of thalamocortical cultures with botulinum toxin C 1 , which specifically cleaves syntaxin 1A , decreased SERT function . ^^^ In addition , co immunoprecipitation experiments showed that SERT and syntaxin 1A form a protein complex in these neurons . ^^^

The present data show that the SNARE protein syntaxin 1A binds the N terminal tail of SERT , and this interaction regulates two SERT conducting states . ^^^ These two SERT conducting states are present endogenously in thalamocortical neurons , act to depolarize the membrane potential , and are modulated by molecules that disrupt SERT and syntaxin 1A interactions . ^^^
