Mutations in the three largest subunits of yeast RNA polymerase 2 ( RPB 1 , RPB 2 , and RPB 3 ) were investigated for their effects on RNA polymerase 2 structure and assembly . ^^^ The time course of assembly of RPB 1 , RPB 2 , and RPB 3 was investigated in wild type cells and subsequently in mutant cells . ^^^ The RPB 2 plus RPB 3 subcomplexes accumulated in all RPB 1 assembly mutants at the nonpermissive temperature but not in an RPB 2 or RPB 3 assembly mutant . ^^^ These data indicate that RPB 2 and RPB 3 form a complex that subsequently interacts with RPB 1 during the assembly of RNA polymerase II . . ^^^

A sensitive phenotypic assay has been used to identify mutations affecting transcription initiation in the genes encoding the two large subunits of Saccharomyces cerevisiae RNA polymerase 2 ( RPB 1 and RPB 2 ) . ^^^ The rpb 1 and rpb 2 mutations alter the ratio of transcripts initiated at two adjacent start sites of a delta insertion promoter . ^^^ Of a large number of rpb 1 and rpb 2 mutations screened , only a few affect transcription initiation patterns at delta insertion promoters , and these mutations are in close proximity to each other within both RPB 1 and RPB 2 . ^^^

The two large subunits of RNA polymerase 2 , RPB 1 and RPB 2 , contain regions of extensive homology to the two large subunits of Escherichia coli RNA polymerase . ^^^ We investigated whether suppressor genetics could provide evidence for interactions between specific segments of RPB 1 and RPB 2 in Saccharomyces cerevisiae . ^^^ A plasmid shuffle method was used to screen thoroughly for mutations in RPB 2 that suppress a temperature sensitive mutation , rpb 1 1 , which is located in region H of RPB 1 . ^^^ All six RPB 2 mutations that suppress rpb 1 1 were clustered in region 1 of RPB 2 . ^^^ The location of these mutations and the observation that they were allele specific for suppression of rpb 1 1 suggests an interaction between region H of RPB 1 and region 1 of RPB 2 . ^^^
