Mutations in the three largest subunits of yeast RNA polymerase 2 ( RPB 1 , RPB 2 , and RPB 3 ) were investigated for their effects on RNA polymerase 2 structure and assembly . ^^^ The time course of assembly of RPB 1 , RPB 2 , and RPB 3 was investigated in wild type cells and subsequently in mutant cells . ^^^ Glycerol gradient fractionation of extracts of cells pulse labeled for various times revealed that a subcomplex of RPB 2 and RPB 3 appears soon after subunit synthesis and can be chased into fully assembled enzyme . ^^^ The RPB 2 plus RPB 3 subcomplexes accumulated in all RPB 1 assembly mutants at the nonpermissive temperature but not in an RPB 2 or RPB 3 assembly mutant . ^^^ These data indicate that RPB 2 and RPB 3 form a complex that subsequently interacts with RPB 1 during the assembly of RNA polymerase II . . ^^^

Far Western blot analysis of S . pombe RNA polymerase 2 using 32P labelled recombinant Rpb 5 fused to glutathione S transferase ( GST ) as a probe , indicated that Rpb 5 binds strongly to membrane immobilized Rpb 1 , Rpb 2 and Rpb 3 and weakly to Rpb 5 and a 15 kDa subunit ( Rpb 8 or Rpb 11 ) . ^^^ In agreement with this result , the 32P labelled Rpb 3 probe showed a strong binding signal against Rpb 5 in addition to Rpb 1 and Rpb 2 . ^^^

After stepwise dissociation of the Rpb 1 and Rpb 3 tagged RNA polymerases fixed on Ni2+ resin by increasing concentrations of urea or guanidium hydrochloride , Rpb 2 Rpb3 Rpb 11 or Rpb 2 Rpb3 Rpb 11 Rpb10 complexes were obtained . ^^^ Since the complex consisting of Rpb 2 , Rpb 3 , and Rpb 11 can not be dissociated even after treatment with 6 M urea buffer , we propose that this complex represents a core subassembly of the RNA polymerase 2 , analogous to the alpha2beta complex in the assembly of Escherichia coli RNA polymerase . ^^^
