Phagocytosis by CD 14 positive THP 1 cells was attenuated by phosphatidylinositol 3 inhibitors LY 294002 and wortmannin and experiments using transfected CHO cells showed substantial accumulation of phosphatidylinositol 3 , 4 , 5 trisphosphate at the BCG attachment site in CHO cells expressing CD 14 and TLR 2 suggesting that bacteria bind to CD 14 and use TLR 2 to initiate a PI3K signaling pathway . ^^^ These findings reveal that mycobacteria promote their uptake through a process of `` inside out ' ' signaling involving CD 14 , TLR 2 , PI3K , and cytohesin 1 . ^^^ |

TLR 2 transmodulates monocyte adhesion and transmigration via Rac 1 and PI3K mediated inside out signaling in response to Porphyromonas gingivalis fimbriae . ^^^ Fimbriae induced CD11b / CD18 dependent adhesion of human monocytes or mouse macrophages to endothelial receptor ICAM 1 ; these activities were inhibited by TLR 2 blockade or deficiency or by pharmacological inhibitors of PI3K . ^^^ Thus , P . gingivalis fimbriae activate a novel proadhesive pathway in human monocytes , involving TLR 2 , Rac 1 , PI3K , and CD11b / CD18 , which may constitute a mechanistic basis linking P . gingivalis to inflammatory atherosclerotic processes . . ^^^ |
