Of all nine PAX genes investigated , only PAX 5 and PAX 6 were consistently expressed in most medulloblastomas ( 70 and 78 % of all cases , respectively ) , as were the genes EN 1 ( 57 % ) and EN 2 ( 78 % ) . ^^^ EN 1 , EN 2 , and PAX 6 genes were also expressed in normal cerebellar tissue , and their expression in medulloblastoma is consistent with the hypothesis that this tumor originates in the external granular layer of the developing cerebellum . ^^^ |

The homeobox containing Engrailed ( En 1 ) product down regulates the expression of Pax 6 through a DNA binding independent mechanism . ^^^ By in situ hybridization of quail neuroretinas , we observed that Engrailed ( En 1 ) is expressed both in the ganglionic and the amacrine cell layers , similarly to Pax 6 . ^^^ Because we observed a decrease of Pax 6 expression in the neuroretina of hatched animals , we studied the effect of the chicken En 1 and En 2 proteins on Pax 6 expression . ^^^ En 1 and to some extent En 2 were able to repress the basal and the p46Pax 6 activated transcription from the two Pax 6 promoters . ^^^ Infection of retinal pigmented epithelium by a virus encoding the En 1 protein repressed the endogenous Pax 6 , and a similar effect was observed with a homeodomain deleted En 1 . ^^^ |

Pax 6 defines the di mesencephalic boundary by repressing En 1 and Pax 2 . ^^^ In the early stage of neural development , Pax 6 is expressed in the prosencephalon , while En 1 and Pax 2 are expressed in the mesencephalon . ^^^ Pax 6 repressed En 1 , Pax 2 and other tectum ( mesencephalon ) related genes such as En 2 , Pax 5 , Pax 7 , but induced Tcf 4 , a diencephalon marker gene . ^^^ To know how Pax 6 represses En 1 and Pax 2 , we ectopically expressed a dominant active or negative form of Pax 6 . ^^^ The results of misexpression experiments , together with normal expression patterns of Pax 6 , En 1 and Pax 2 , suggest that repressive interaction between Pax 6 and En1 / Pax2 defines the di mesencephalic boundary . . ^^^ |
