Here we show that cdk8 / cyclin C can regulate transcription by targeting the cdk7 / cyclin H subunits of the general transcription initiation factor IIH ( TFIIH ) . cdk 8 phosphorylates mammalian cyclin H in the vicinity of its functionally unique amino terminal and carboxy terminal alpha helical domains . ^^^

Other proteins identified in IGCs include transcription factors , the large subunit of RNA polymerase 2 ( Pol 2 ) , and cyclin dependent kinase 8 ( cdk 8 ) . cdk 8 is the kinase partner of cyclin C and a component of the mediator complex , associated with the Pol 2 holoenzyme . cdk 8 and cyclin C can regulate transcription via phosphorylation of cyclin H and the carboxy terminal domain of Pol 2 . ^^^

RESULTS : Cyclin H was overexpressed in all tumours , cyclin C in 88 % , cyclin B 1 in 58 % , cyclin A in 83 % , cyclin D 3 in 36 % , cyclin E in 25 % , and cyclin D 1 in 11 % of the tumours . ^^^ Extra gene copies of cyclin A were seen in 6 . 2 % of the tumours , cyclin B 1 in 9 % , cyclin C in 26 . 9 % , cyclin D 1 in 55 % , cyclin D 3 in 20 . 5 % , cyclin E in 19 . 1 % , and cyclin H in 5 . 1 % . ^^^

RESULT : Of the 80 breast tumour samples available for cyclin A gene amplification analyses , 26 . 7 % ( 23 / 80 ) was defined to have cyclin A gene amplification . 37 . 2 % ( 32 / 79 ) had cyclin B 1 gene amplification , 82 . 6 % ( 71 / 82 ) of the samples harboured amplification of cyclin C gene , 74 . 4 % ( 64 / 82 ) had cyclin D 1 gene amplification , 41 . 9 % ( 36 / 86 ) had cyclin D 3 gene amplification , 29 . 1 % ( 25 / 81 ) of the patients had cyclin E gene amplification and 9 . 3 % ( 8 / 86 ) of the samples showed amplification of the cyclin H gene . ^^^
