Interaction of SP 100 with HP 1 proteins : a link between the promyelocytic leukemia associated nuclear bodies and the chromatin compartment . ^^^ Here , we show that SP 100 binds to members of the heterochromatin protein 1 ( HP 1 ) families of non histone chromosomal proteins . ^^^ Both HP 1 and SP 100 HMG concentrate in the PML / SP100 NBs , and overexpression of SP 100 leads to enhanced accumulation of endogenous HP 1 in these structures . ^^^ When bound to a promoter , SP 100 , SP 100 HMG and HP 1 behave as transcriptional repressors in transfected mammalian cells . ^^^

In this report , we have reconstituted and characterized the HP 1 KAP 1 interaction using purified proteins and have compared KAP 1 to three other known HP 1 binding proteins : SP 100 , lamin B receptor ( LBR ) , and the p 150 subunit from chromatin assembly factor ( CAF 1 p 150 ) . ^^^ The use of a spectrum of amino acid substitutions in the human HP1alpha CSD revealed a strong correlation between CSD mediated repression and binding to KAP 1 , CAF 1 p 150 , and SP 100 but not LBR . ^^^

In this study , we found that EBNA LP interacts with the promyelocytic leukemia nuclear body ( PML NB ) associated protein Sp 100 and displaces Sp 100 and heterochromatin protein 1alpha ( HP1alpha ) from PML NBs . ^^^ Overexpression of Sp 100 lacking the N terminal PML NB targeting domain , but not a mutant form of Sp 100 lacking the HP1alpha interaction domain , was sufficient to coactivate EBNA 2 in a gene specific manner independent of EBNA LP . ^^^
