We show here that RFX 1 belongs to a previously unidentified family of DNA binding proteins of which we have cloned three members , RFX 1 , RFX 2 , and RFX 3 , from humans and mice . ^^^ They have very similar DNA binding specificities and heterodimerize both in vitro and in vivo . mRNA levels for all three genes , particularly RFX 2 , are elevated in testis . ^^^ In other cell lines and tissues , RFX mRNA levels are variable , particularly for RFX 2 and RFX 3 . ^^^ |

Here , we demonstrate by yeast one hybrid screening that RFX 2 protein specifically binds to this cis element . ^^^ RFX 2 belongs to the RFX DNA binding protein family , the biological role of which remains obscure . ^^^ Using an electrophoretic mobility shift assay , we further show that RFX 1 , RFX 2 , and RFX 3 homodimers and heterodimers specifically bind to the cis element of the IL 5Ralpha promoter . ^^^ The mRNA expression of RFX 1 , RFX 2 , and RFX 3 was detected ubiquitously , but in transient transfection assays , multimerized RFX binding sites in front of a basal promoter efficiently functioned in a tissue and lineage specific manner . ^^^ |

Regulatory factor X 2 ( RFX 2 ) binds to the H1t / TE1 promoter element and activates transcription of the testis specific histone H1t gene . ^^^ We purified an 85 kDa protein from rat testis nuclear extracts using the TE 1 subelement as an affinity chromatography probe and analysis revealed that the protein was RFX 2 . ^^^ Polyclonal antibodies directed against RFX 2 supershift the low mobility testis nuclear protein complex formed in electrophoretic mobility shift assays ( EMSA ) . ^^^ RFX 2 derived from primary spermatocytes , where the transcription factor is relatively abundant , binds with high affinity to the TE 1 element . ^^^ Coexpression of RFX 2 together with an H1t promoter / reporter vector activates the H1t promoter in a cultured GC 2spd germinal cell line , but mutation of either the TE 1 subelement or the TE 2 subelements represses activity . ^^^ |

RFX 2 is a potential transcriptional regulatory factor for histone H1t and other genes expressed during the meiotic phase of spermatogenesis . ^^^ The testis specific binding complex contains RFX 2 , probably as a homodimer . ^^^ Western blots confirmed that RFX 2 expression is greatly enhanced in adult testis and that RFX 2 is equally prominent in highly enriched populations of late pachytene spermatocytes and round spermatids . ^^^ Immunohistochemistry carried out on mouse testis showed that RFX 2 is strongly expressed in pachytene spermatocytes , remains high in early round spermatids , and declines only in advance of nuclear condensation . ^^^ RFX specific band complexes were also identified for both the mouse lamin C 2 and Sgy promoters , using either testis nuclear extracts or in vitro synthesized RFX 2 . ^^^ |
