Mutations in the histone fold domain of the TAF 12 gene show synthetic lethality with the TAF 1 gene lacking the TAF N terminal domain ( TAND ) by different mechanisms from those in the SPT 15 gene encoding the TATA box binding protein ( TBP ) . ^^^ Interestingly , a set of genes affected in the taf 1 DeltaTAND mutant is similarly affected in the taf 12 HFD mutants but not in the nsl mutants of TBP . ^^^ |

Our analysis also indicates that TAF 5 , TAF 6 , TAF 9 , and TAF 12 play key roles in stability of the complex , whereas TBP , TAF 1 , TAF 2 , and TAF 11 contribute very little to complex stability . ^^^ Based on our results , we propose that holo TFIID comprises a stable core subcomplex containing TAF 4 , TAF 5 , TAF 6 , TAF 9 , and TAF 12 decorated with peripheral subunits TAF 1 , TAF 2 , TAF 11 , and TBP . ^^^ |

Coimmunoprecipitation assays showed that TAFII 80 interacted with TAFII 250 , TAFII 31 , TAFII 20 , and TBP , but not with TAFII 55 . ^^^ The N terminus of TAFII 80 ( residues 1 100 ) interacts with both TAFII 31 and TAFII 20 , while two C terminal regions are involved , respectively , in interactions with TAFII 250 and TFIIF alpha ( RAP 74 ) ( residues 203 276 ) and with TBP and TFIIE alpha ( residues 377 505 ) . ^^^ |
