The N terminus of TAFII 80 ( residues 1 100 ) interacts with both TAFII 31 and TAFII 20 , while two C terminal regions are involved , respectively , in interactions with TAFII 250 and TFIIF alpha ( RAP 74 ) ( residues 203 276 ) and with TBP and TFIIE alpha ( residues 377 505 ) . ^^^

TAFII 250 is a bipartite protein kinase that phosphorylates the base transcription factor RAP 74 . ^^^ Here , we report that purified recombinant TAFII 250 is a protein serine kinase that selectively phosphotylates RAP 74 but not other basal transcription factors or common phosphoacceptor proteins . ^^^ However , both the N and C terminal kinase domains of TAFII 250 are required for efficient transphosphorylation of RAP 74 on serine residues . ^^^ These findings suggest that the targeted phosphorylation of RAP 74 by TAFII 250 may provide a mechanism for signaling between components within the initiation complex to regulate transcription . . ^^^

Rb inhibits the autophosphorylation of TAFII 250 as well as its phosphorylation of the RAP 74 subunit of TFIIF in a dose responsive manner . ^^^ Our results suggest a novel mechanism of transcriptional regulation by Rb , involving direct interaction with TAFII 250 and inhibition of its ability to phosphorylate itself , RAP 74 , and possibly other targets . . ^^^
