The GTPase activating protein , p120GAP , contains an amino acid sequence motif called the Ca2+ dependent lipid binding domain ( CaLB ) which mediates a protein protein interaction between p120GAP and annexin 6 and also binds to negatively charged phospholipids . ^^^ Because membrane association of p120GAP is important for the regulation of p 21 Ras activity , we have studied the roles played by Ca2+ , phospholipids and annexin 6 in the membrane association of p120GAP . ^^^ Thus , annexin 6 may simultaneously mediate an interaction with p120GAP and also an interaction with membrane phospholipids . ^^^ This may in part explain the mechanism by which p120GAP associates with membranes in response to Ca2+ elevation and suggests the potential importance of annexin 6 in the regulation of p 21 Ras and the role CaLB domains may play in the specific recognition of cellular membranes . . ^^^

Identification of a novel protein complex containing annexin 6 , Fyn , Pyk 2 , and the p 120 ( GAP ) C 2 domain . p 120 ( GAP ) ( RasGAP ) has been proposed to function as both an inhibitor and effector of Ras . ^^^ Specifically , three proteins have been isolated in a complex with the C 2 domain of RasGAP ; these are the Ca ( 2+ ) dependent lipid binding protein annexin 6 ( p 70 ) and two previously unidentified proteins , p 55 and p 120 . ^^^ Finally , co immunoprecipitation studies in Rat 1 fibroblasts confirm that Fyn , Pyk 2 , annexin 6 and RasGAP can form a protein complex in mammalian cells . . ^^^

Annexin A 6 is a calcium dependent membrane binding protein that interacts with signalling proteins , including the GTPase activating protein p120GAP , one of the most important inactivators of Ras . ^^^ Annexin A 6 stimulates the membrane recruitment of p120GAP to modulate Ras and Raf 1 activity . ^^^ Since we have demonstrated that annexin A 6 inhibits EGF and TPA induced Ras signalling , we investigated whether modulation of Ras activity by annexin A 6 was mediated via altered subcellular localization of p120GAP . ^^^ Annexin A 6 promoted membrane binding of p120GAP in vitro , and plasma membrane targeting of p120GAP in living cells , both in a Ca ( 2+ ) dependent manner , which is consistent with annexin A 6 promoting the Ca ( 2+ ) dependent assembly of p120GAP Ras at the plasma membrane . ^^^ Importantly , the enhancement of Ras activation following RNAi mediated reduction in p120GAP levels was more marked in annexin A 6 expressing A 431 cells than controls , indicating that the effect of annexin A 6 on Ras was mediated via p120GAP . ^^^
