The human dok 2 cDNA encodes a 412 amino acid protein with a predicted N terminal pleckstrin homology domain as well as several other features of a signaling molecule , including 13 potential tyrosine phosphorylation sites , six PXXP motifs , and the ability to bind to p 120 ( RasGAP ) . ^^^ Dok 2 was shown to be 35 % identical to p 62 ( dok 1 ) , a recently identified RasGAP binding protein from CML cells , and analysis of the expressed sequence tag data base revealed the presence of at least four additional proteins containing a Dok homology sequence motif . ^^^
Interacting proteins: O60496 and P20936 Pubmed SVM Score :0.0
Dok 4 and dok 5 do not associate with rasGAP or Nck , in contrast to p62dok and dok 2 . ^^^
Interacting proteins: O60496 and P20936 Pubmed SVM Score :0.0
This signal involves the SH 2 domain containing inositol phosphatase ( SHIP ) ; the adaptor molecules Dok 2 , Dok 1 and Shc ; and Ras GTPase activating protein RasGAP . ^^^
Interacting proteins: O60496 and P20936 Pubmed SVM Score :0.0
Upon phosphorylation , Dok 1 binds to SHIP and both Dok 1 and Dok 2 recruit RasGAP , which mediates the inhibition of the Ras / MAPK pathways . ^^^
Interacting proteins: O60496 and P20936 Pubmed SVM Score :0.0
Members of this family such as p62dok ( Dok 1 ) and p56dok 2 ( Dok 2 ) associate with the p 120 rasGTPase activating protein ( rasGAP ) upon phosphorylation by p210bcr / abl as well as receptor and nonreceptor tyrosine kinases . ^^^
