Tissue levels of mRNA for ubiquitin and the 20S proteasome subunits HC 3 , HC 5 , HC 7 , and HC 9 were determined by dot blot analysis . ^^^

Of these multiple components , the nucleotide sequences of five major subunits ( named HC 2 , HC 3 , HC 5 , HC 8 and HC 9 ) of human proteasomes have been determined from recombinant cDNA clones by screening a human HepG 2 hepatoblastoma cell cDNA library with rat proteasome cDNAs isolated previously as probes . ^^^

C . diphtheriae mutant C7hm723 produced siderophore and toxin constitutively under low iron and high iron conditions , but in mutants HC 1 , HC 3 , HC 4 , and HC 5 their synthesis was partially repressed under high iron conditions . ^^^ The phenotypes of HC 1 , HC 3 , HC 4 , and HC 5 are consistent with their severe defects in iron uptake , but the phenotype of C7hm723 is more likely to be explained by inactivation of the repressor for the iron regulon of C . diphtheriae . . ^^^

These results differed from previous findings that the genes for the alpha type HC 3 and beta type HC 5 subunits of the human proteasome have a TATA less promoter and that two or three GC boxes function as the promoter sequences ( Tamura , T . et al . ( 1994 ) J . ^^^

Isolation and characterization of alpha type HC 3 and beta type HC 5 subunit genes of human proteasomes . ^^^ To understand the molecular basis of the proteasome genes , we isolated and characterized two human proteasome genes econding the alpha type HC 3 and beta type HC 5 subunit . ^^^ The functional genes for HC 3 and HC 5 are similar in being approximately 15 kb in length , but differ in having exon numbers of 9 and 6 , respectively . ^^^ The HC 5 gene does not have this silence element , and its promoter activity is five to ten times that of HC 3 . ^^^ These results show that the human proteasomal HC 3 and HC 5 genes differ not only in their genomic structures , such as their numbers of exons and their exon intron organizations , but also in the mechanisms regulating their transcription , suggesting that they diverged at an early stage of evolution . . ^^^

IRP 1 and IRP 6 were repressed under high iron conditions in wild type C . diphtheriae C 7 ( beta ) , but they were expressed constitutively in C 7 ( beta ) mutant strains HC 1 , HC 3 , HC 4 , and HC 5 , which were shown previously to be defective in corynebactin dependent iron uptake . ^^^ A clone of the wild type irp 6 operon ( pCM6ABC ) complemented the constitutive corynebactin production phenotype of HC 1 , HC 4 , and HC 5 but not of HC 3 , whereas a clone of the wild type irp 1 operon failed to complement any of these strains . ^^^
