| Rab 5 and Rin 1 , a multifunctional protein with a Rab 5 guanine nucleotide exchange factor domain , have been shown to regulate EGFR endocytosis ( Barbieri et al . , 2000 ; Tall et al . , 2001 ) . ^^^ In this study , we demonstrate that overexpression of both dominant negative Rab 5 ( Rab 5 : S34N ) and full length Rin 1 selectively block EGF activation of the Raf Erk1 / 2 kinase pathway and EGF stimulated incorporation of [ 3H ] thymidine into DNA without affecting the activity of JN and p 38 kinase pathways . ^^^ Expression of Rab 5 : S34N and Rin 1 also block EGF induction of cyclin D 1 transcription . ^^^ In contrast , expression of Rin 1 : delta , a natural splice variant of Rin 1 lacking 47 amino acids in the Vps9p domain or Rab 5 , increase both activation of Raf Erk1 / 2 and cyclin D 1 transcription in response to EGF . ^^^ |

| Rin 1 is a multifunctional protein containing several domains , including Ras binding and Rab 5 GEF domains . ^^^ Here , we show that insulin receptor mediated endocystosis and fluid phase insulin stimulated endocytosis are enhanced in cells expressing the Rin 1 : wild type and the Rin 1 : C deletion mutant , which contain both the Rab 5 GEF and GTP bound Ras binding domains . ^^^ |
