In contrast to Rab 4 , which also leaves the low density microsomes in response to insulin , Rab 5 was not found in Glut 4 containing vesicles purified by immunoadsorption on antibodies to Glut 4 . ^^^ When adipocytes were treated with wortmannin , an inhibitor of phosphatidylinositol 3 kinase , the effect of insulin on Rab 5 movement was not affected , whereas the insulin induced movements of Rab 4 and Glut 4 were abolished . ^^^ As a whole , our results suggest that Rab 5 is not involved in insulin stimulated Glut 4 exocytosis . ^^^ |

Insulin can regulate GLUT 4 internalization by signaling to Rab 5 and the motor protein dynein . ^^^ Microinjection of anti Rab 5 or anti dynein antibody into 3T3 L 1 adipocytes increased the basal level of surface GLUT 4 , did not change the insulin stimulated surface GLUT 4 level , and inhibited GLUT 4 internalization after the removal of insulin . ^^^ In conclusion , these data indicate that insulin signaling inhibits Rab 5 activity and the interaction of dynein with microtubules in a PI 3 kinase dependent manner , and that these effects may inhibit the rate of GLUT 4 internalization . ^^^ As such , our results present a previously uncharacterized insulin signaling pathway involving Rab 5 , the motor protein dynein , and the cytoskeleton to regulate directional GLUT 4 movement , facilitating GLUT 4 distribution to the cell surface . . ^^^ |

To evaluate the role of the small rab GTP binding proteins in glucose transporter trafficking , we have heterologously co expressed rab 4 or rab 5 and GLUT 4 or GLUT 1 glucose transporters in Xenopus oocytes . ^^^ Co injection of rab 4 and GLUT 4 cRNAs resulted in a dose dependent decrease in glucose transport ; this effect was specific for rab 4 , since co injection of an inactive rab 4 mutant or rab 5 cRNA did not have any effect on glucose transport . ^^^ |
