| Interaction with general transcription factor IIF ( TFIIF ) is required for the suppression of activated transcription by RPB 5 mediating protein ( RMP ) . ^^^ RMP was reported to regulate transcription via competing with HBx to bind the general transcription factor IIB ( TFIIB ) and interacting with RPB 5 subunit of RNA polymerase 2 as a corepressor of transcription regulator . ^^^ The interaction between RMP and TFIIF may be an additional passway for RMP to regulate the transcription , or alternatively TFIIF may cooperate with RPB 5 and TFIIB for the corepressor function of RMP . . ^^^ |

| By far Western blot screening , we cloned a novel gene encoding a 508 amino acid residue RPB 5 binding protein from a HepG 2 cDNA library and designated it RPB 5 mediating protein ( RMP ) . ^^^ Bacterially expressed recombinant RMP strongly bound RPB 5 but neither HBx nor TATA binding protein in vitro . ^^^ Endogenous RMP was immunologically detected interacting with assembled RPB 5 in RNA polymerase in mammalian cells . ^^^ The central part of RMP is responsible for RPB 5 binding , and the RMP binding region covers both the TFIIB and HBx binding sites of RPB 5 . ^^^ Overexpression of RMP , but not mutant RMP lacking the RPB 5 binding region , inhibited HBx transactivation of reporters with different HBx responsive cis elements in transiently transfected cells . ^^^ |
