Far Western blot analysis of S . pombe RNA polymerase 2 using 32P labelled recombinant Rpb 5 fused to glutathione S transferase ( GST ) as a probe , indicated that Rpb 5 binds strongly to membrane immobilized Rpb 1 , Rpb 2 and Rpb 3 and weakly to Rpb 5 and a 15 kDa subunit ( Rpb 8 or Rpb 11 ) . ^^^ In agreement with this result , the 32P labelled Rpb 3 probe showed a strong binding signal against Rpb 5 in addition to Rpb 1 and Rpb 2 . ^^^ CONCLUSION : Rpb 3 and Rpb 5 , the putative subunits of RNA polymerase 2 , associate each other to form binary complexes . ^^^

Previously we indicated that Rpb 3 also plays an essential role in subunit assembly because it interacts with at least four subunits , two large subunits ( Rpb 1 and Rpb 2 ) and two medium sized subunits ( Rpb 3 and Rpb 5 ) ( 1 ) , and it constitutes a core subassembly consisting of Rpb 2 , Rpb 3 , and Rpb 11 ( 2 ) . ^^^ By making a set of Rpb 3 deletion derivatives , we carried out mapping of the Rpb 5 and Rpb 11 contact sites on Rpb 3 . ^^^ By far Western blot and GST pull down assays , we found that the amino acid sequence between residues 105 263 of Rpb 3 is involved in binding Rpb 5 , and the sequence between residues 105 297 is required for binding Rpb 11 . ^^^ Although the Rpb 5 and Rpb 11 contact sites on Rpb 3 overlap each other , both subunits are able to associate with Rpb 3 simultaneously . ^^^ The binding of Rpb 5 stabilizes the Rpb 3 Rpb11 heterodimer . . ^^^
