T cell receptor induced activation of phospholipase C gamma 1 depends on a sequence independent function of the P 1 region of SLP 76 . ^^^ T cell receptor ( TCR ) induced activation of phospholipase C gamma 1 ( PLC gamma 1 ) depends on three features of SLP 76 : the N terminal tyrosine phosphorylation sites , the Gads binding site , and an intervening sequence , denoted the P 1 region , which binds to the SH 3 domain of PLC gamma 1 ( SH 3 ( PLC ) ) via a low affinity interaction . ^^^ Despite extensive research , the mechanism whereby SLP 76 regulates PLC gamma 1 remains uncertain . ^^^ To better understand the contribution of the P 1 region to PLC gamma 1 activation , we mapped the PLC gamma 1 binding site within the region , and created a SLP 76 mutant that fails to bind SH 3 ( PLC ) , but is fully functional , mediating TCR induced phosphorylation of PLC gamma 1 at tyrosine 783 , calcium flux , and nuclear factor of activated T cells activation . ^^^ Our results demonstrate that SLP 76 need not interact with SH 3 ( PLC ) to activate PLC gamma 1 , and further suggest that the P 1 region of SLP 76 serves a structural role that is sequence independent and is not directly related to protein protein interactions . . ^^^

Following T cell receptor ligation , PLCgamma 1 interacts through its SH 2 and SH 3 domains with the adaptors LAT and SLP 76 , respectively , to form a multiprotein signaling complex that leads to activation of PLCgamma 1 by Syk tyrosine kinases . ^^^ To identify the binding site for PLCgamma 1 in SLP 76 , we used isothermal titration calorimetry to measure affinities for the interaction of PLCgamma 1 SH3 with a set of overlapping peptides spanning the central proline rich region of SLP 76 . ^^^ PLCgamma 1 SH3 bound with high specificity to the SLP 76 motif 186PPVPPQRP193 , which represents the minimal binding site . ^^^ In addition , Pro 193 of SLP 76 assists in positioning Arg 192 into the compass pocket of PLCgamma 1 SH3 , which coordinates the compass residue through an unusual aspartate . ^^^ The PLCgamma 1 SH3 / SLP 76 structure provides insights into ligand binding by SH 3 domains related to PLCgamma 1 SH3 , as well as into recognition by PLCgamma 1 of signaling partners other than SLP 76 . . ^^^
