| The thrombin induced IL 8 / CXCL8 release was attenuated by D phenylalanyl L prolyl L arginine chloromethyl ketone ( a thrombin inhibitor ) , U 73122 ( a phosphoinositide phospholipase C inhibitor ) , Ro 32 0432 ( a protein kinsase C alpha ( PKC alpha ) inhibitor ) , an NF kappaB inhibitor peptide , and Bay 117082 ( an IkappaB phosphorylation inhibitor ) . ^^^ These results show for the first time that thrombin , acting through PAR 1 and PAR 4 , activates the phosphoinositide phospholipase C / PKC alpha / c Src / IKK alphabeta signaling pathway to induce NF kappaB activation , which in turn induces IL 8 / CXCL8 expression and release in human lung EC . . ^^^ |

| The TNF alpha induced , but not the TPA induced , COX 2 promoter activity was inhibited by phospholipase C gamma 2 mutants , and the COX 2 promoter activity induced by either agent was attenuated by dominant negative mutants of PKC alpha , NF kappaB inducing kinase , or 1 kappaB ( inhibitory protein that dissociates from NF kappaB ) kinase ( IKK ) 1 or 2 . ^^^ These results suggest that , in NCI H 292 epithelial cells , TNF alpha might activate phospholipase C gamma 2 via an upstream tyrosine kinase to induce activation of PKC alpha and protein tyrosine kinase , resulting in the activation of NF kappaB inducing kinase and IKK1 / 2 , and NF kappaB in the COX 2 promoter , then initiation of COX 2 expression and PGE 2 release . . ^^^ |
