FER kinase activation of Stat 3 is determined by the N terminal sequence . p 94 ( fer ) and p 51 ( ferT ) are two tyrosine kinases that share identical SH 2 and kinase domains but differ in their N terminal regions . ^^^ When combined with immunoprecipitation analysis , this assay showed that p 94 ( fer ) can lead to the tyrosine phosphorylation and activation of Stat 3 but not of Stat 1 or Stat 2 . ^^^ However , modified forms of p 94 ( fer ) , which are constitutively nuclear , could also lead to the phosphorylation of Stat 3 . ^^^ Endogenous Stat 3 and p 94 ( fer ) co immunoprecipitated with each other , thus proving the interaction of these two proteins in vivo . ^^^ Unlike p 94 ( fer ) , p 51 ( ferT ) did not induce the phosphorylation of Stat 3 but led to the phosphorylation of other nuclear proteins . ^^^ |

Gamma interferon down regulates Fer and induces its association with inactive Stat 3 in colon carcinoma cells . ^^^ Up and down regulation of Fer was paralleled by the activation and subsequent deactivation of Stat 3 , which is a potent oncogene and a putative substrate of the tyrosine kinase Fer . ^^^ Moreover , IFN gamma induced the association of Fer and Stat 3 and the newly formed complex was most stable at the down regulated states of the two proteins . ^^^ Thus , Fer and Stat 3 are two proliferation promoting factors whose down regulation could contribute to the cytostatic activity of IFN gamma in colon carcinoma cells . . ^^^ |
