Pendulin is a new member of a superfamily of proteins which contains Armadillo ( Arm ) repeats and displays extensive sequence similarities with the Srp 1 protein from yeast , with RAG 1 interacting proteins from humans , and with the importin protein from Xenopus . ^^^ |

RAG 1 interacts with the repeated amino acid motif of the human homologue of the yeast protein SRP 1 . ^^^ After cotransfection of the genes encoding RAG 1 and human SRP 1 into 293T cells , a stable complex was evident . ^^^ Deletion analysis indicated that the region of the SRP 1 protein interacting with RAG 1 involved four repeats . ^^^ The domain of RAG 1 that associates with SRP 1 mapped N terminal to the zinc finger domain . ^^^ Because this region of RAG 1 is not required for recombination and SRP 1 appears to be bound to the nuclear envelope , we suggest that this interaction helps to localize RAG 1 . . ^^^ |

Despite the fact that BIIa is important for the interaction of RAG 1 with the nuclear localization factor Srp 1 , it does not appear to enhance recombination by facilitating nuclear transport of RAG 1 . ^^^ |

Nucleolar / nuclear localization of RAG 1 is mediated by four basic domains , which are the binding sites for the nuclear transport proteins SRP 1 and RCH 1 , and by a nuclear localization signal ( NLS ) in the fifth basic domain . ^^^ |
