| The effect on VEGFR 2 function was due to direct binding and sequestration of VEGF 165 by ADAMTS 1 . ^^^ The interaction required the heparin binding domain of the growth factor , because VEGF 121 failed to bind to ADAMTS 1 . ^^^ Structure / function analysis with independent ADAMTS 1 domains indicated that binding to VEGF 165 was mediated by the carboxyl terminal ( CT ) region . ^^^ ADAMTS 1 and VEGF 165 were also found in association in tumor extracts . ^^^ These findings provide a mechanism for the anti angiogenic activity of ADAMTS 1 and describe a novel modulator of VEGF bioavailability . . ^^^ |

| The purpose of this study was to investigate the expression of ADAMTS 1 in endothelial cells and in a mouse model of ischemia induced retinal neovascularization and to study the regulation of ADAMTS 1 expression in endothelial cells by vascular endothelial growth factor ( VEGF ) . ^^^ RESULTS : ADAMTS 1 mRNA and protein levels were increased in a mouse model of ischemia induced retinal neovascularization , and VEGF induced time and dose dependent increases in ADAMTS 1 mRNA and protein expression in endothelial cells . ^^^ VEGF upregulation of ADAMTS 1 expression was completely abolished by the inhibition of protein kinase C by calphostin C and largely blocked by the specific inhibition of PKCbeta . ^^^ CONCLUSIONS : These results indicate that VEGF significantly induces ADAMTS 1 expression in endothelial cells in a PKC dependent fashion . ^^^ ADAMTS 1 expression is also increased , along with VEGF expression , in vivo in ischemia induced retinal neovascularization . ^^^ |
