It has been found that , ( 1 ) gp 96 is one of the major peptide binding proteins in the ER , ( 2 ) gp 96 interacts specifically with receptors including CD 91 and possibly toll like receptors ( TLRs ) , on the surface of professional antigen presenting cells ( APCs ) , ( 3 ) interaction with APCs leads to re presentation of gp 96 chaperoned peptides to the major histocompatibility complex ( MHC ) molecules of APCs , ( 4 ) direct access of gp 96 to APCs triggers functional activation of APCs . ^^^

The CD 91 molecule ( also called alpha 2 macroglobulin receptor or the low density lipoprotein related protein ) is shown here to be a cell surface receptor for the heat shock protein gp 96 . ^^^ CD 91 : a receptor for heat shock protein gp 96 . ^^^ CD 91 binds gp 96 directly , rather than through another ligand for CD 91 . ^^^ The previously known CD 91 ligand , alpha 2 macroglobulin , inhibits re presentation of gp 96 chaperoned antigenic peptides by macrophages , as do antibodies to CD 91 . ^^^ As gp 96 is exclusively intracellular and is released as a result of necrotic but not apoptotic cell death , we propose that CD 91 acts as a sensor for necrotic cell death . . ^^^
