| Here , we provide the first evidence that direct tyrosine phosphorylation by Src family protein tyrosine kinases ( PTKs ) is a novel mechanism for modulating TRPC 6 channel activity . ^^^ We found that TRPC 6 is tyrosine phosphorylated in COS 7 cells when coexpressed with Fyn , a member of the Src family PTKs . ^^^ This epidermal growth factor induced tyrosine phosphorylation of TRPC 6 was significantly blocked by PP 2 , a specific inhibitor of Src family PTKs , and by a dominant negative form of Fyn , suggesting that the direct phosphorylation of TRPC 6 by Src family PTKs could be caused by physiological stimulation . ^^^ Thus , our findings demonstrated that tyrosine phosphorylation by Src family PTKs is a novel regulatory mechanism of TRPC 6 channel activity . . ^^^ |

| This requirement is unique for TRPC 3 because ( 1 ) mutation of the cognate tyrosines of the closely related TRPC 6 and TRPC 7 had no effect ; ( 2 ) TRPC 6 and TRPC 7 were activated in src , yes , and fyn deficient cells ; and ( 3 ) src , but not yes or fyn , rescued TRPC 3 activation in src , yes , and fyn deficient cells . ^^^ |
