| A novel SH 2 containing phosphatidylinositol 3 , 4 , 5 trisphosphate 5 phosphatase ( SHIP 2 ) is constitutively tyrosine phosphorylated and associated with src homologous and collagen gene ( SHC ) in chronic myelogenous leukemia progenitor cells . ^^^ We describe here the purification and mass spectrometric identification of a 155 kD tyrosine phosphorylated protein associated with src homologous and collagen gene ( SHC ) from p 210 ( bcr / abl ) expressing hematopoietic cells as SHIP 2 , a recently reported , unique SH 2 domain containing protein closely related to phosphatidylinositol polyphosphate 5 phosphatase SHIP . ^^^ Interestingly , SHIP 2 was found to selectively bind to the SH 3 domain of ABL , whereas SHIP 1 selectively binds to the SH 3 domain of Src . ^^^ |

| Src family tyrosine kinases regulate adhesion dependent tyrosine phosphorylation of 5 ' inositol phosphatase SHIP 2 during cell attachment and spreading on collagen 1 . ^^^ In this study , we further characterize the adhesion dependent tyrosine phosphorylation of SHIP 2 and examine the role of Src family tyrosine kinases in the regulation of SHIP 2 function . ^^^ SHIP 2 tyrosine phosphorylation , induced by plating on a collagen 1 coated surface but not by epidermal growth factor or insulin treatment of cells , was completely blocked by small molecule inhibitors of Src family kinases . ^^^ SHIP 2 could be phosphorylated in vitro by recombinant Src kinase and tyrosines 986 987 in the NPXY motif of SHIP 2 appear to be the major sites of phosphorylation for Src both in vitro and in vivo . ^^^ An activated form of Src induced strong tyrosine phosphorylation of SHIP 2 while a dominant negative form decreased collagen 1 dependent SHIP 2 phosphorylation . ^^^ |
