| The enzyme linked immunosorbent assay ( ELISA ) with bacterial heat shock protein 60 ( HSP 60 ) purified from Yersinia enterocolitica ( Ye ) revealed that the antibodies directed against YeHSP 60 existed in sera of patients with primary biliary cirrhosis ( PBC ) . ^^^ The results have suggested that the epitope recognized with anti HSP 60 antibodies in PBC relates to the amino acid sequence of YeHSP 60 molecule as follows : DLGQAKRVVINKDTTIIIDGVGDEAAIQGRLAQIRQQIEEATSDYDKEK . . ^^^ |

| METHODS : Hepatic expression of HSP 60 in frozen liver biopsy specimens from patients with primary sclerosing cholangitis ( PSC ) , primary biliary cirrhosis ( PBC ) , auto immune hepatitis ( AIH ) , obstructive jaundice ( LDO ) , alcoholic liver disease ( ALD ) , and from normal controls was studied by immunohistochemistry using the APAAP technique and confocal laser scanning microscopy . ^^^ RESULTS : Increased expression of HSP 60 was demonstrated in the biliary epithelium of patients with PBC , LDO and , to a lesser extent , with PSC . ^^^ |

| Immunoprecipitation assays with a monoclonal antibody directed against the Hsp 65 kD of mycobacteria and with sera from patients with PBC revealed similar reacting profiles characterized by the precipitation of the overexpressed 65 kD polypeptide from M . gordonae . ^^^ Competitive immunoblotting showed that binding of the monoclonal antibody to the Hsp 65 kD protein was prevented by preincubation with sera from patients with PBC , but not with sera from healthy subjects . ^^^ Furthermore , monoclonal antibody to the Hsp 65 kD protein recognized the main mitochondrial autoantigens of PBC ( PDH E 2 and BCKDH E 2 ) . ^^^ These data indicate the existence of cross reacting epitopes contained on M . gordonae Hsp 65 kD and the main mitochondrial antigens in patients with PBC . . ^^^ |
