Gli 2 is targeted for ubiquitination and degradation by beta TrCP ubiquitin ligase . ^^^ Here we report that beta transducin repeat containing protein ( beta TrCP ) E 3 ubiquitin ligase is required for Gli 2 degradation . beta TrCP 2 directly binds wild type Gli 2 and promotes its ubiquitination . ^^^ Single amino acid substitution in Gli 2 putative binding site inhibits its interaction with beta TrCP 2 , its ubiquitination , and stabilizes the Gli 2 protein . ^^^ These data identify beta TrCP 2 as a pivotal regulator of Gli 2 expression and point to an important role for posttranslational modulation of GLI 2 protein levels in Hh pathway associated human prostate cancer . . ^^^
