In cultured cells , mSu ( fu ) overexpression inhibited transcriptional activation mediated by Sonic hedgehog ( Shh ) , Gli 1 and Gli 2 . ^^^ Co immunoprecipitation of epitope tagged proteins indicated that mSu ( fu ) interacts with Gli 1 , Gli 2 , and Gli 3 , and that the inhibitory effects of mSu ( fu ) on Gli 1 ' s transcriptional activity were mediated through interactions with both amino and carboxy terminal regions of Gli 1 . ^^^ |

HSu ( fu ) was found to repress activity of the zinc finger transcription factor Gli , which mediates Hedgehog signaling in vertebrates , and to physically interact with Gli , Gli 2 and Gli 3 as well as with Slimb , an F box containing protein which , in the fly , suppresses the Hedgehog response , in part by stimulating the degradation of the fly Gli homologue . ^^^ |

This long FU and a much shorter isoform were compared for the ability to regulate GLI 1 and GLI 2 . ^^^ None of the FU isoforms showed any effects on GLI 1 induced transcription but the long form can enhance GLI 2 activity . ^^^ Furthermore , FU is able to enhance the activity of GLI 2 but not of GLI 1 , implicating FU in some aspects of Hedgehog signaling . . ^^^ |
