A study was undertaken to determine whether emphysema and airway secretory cell metaplasia , induced in hamsters by intratracheal treatment with human neutrophil elastase ( HNE ) , could be moderated by pretreatment with human alpha 1 protease inhibitor ( API ) . ^^^ API ( 4 . 9 mg ) was given intratracheally to hamsters 1 h before 0 . 3 mg HNE . ^^^ API given 1 h before HNE moderated the development of bronchial secretory cell metaplasia . ^^^ We calculate that for 50 % protection from emphysema the molar ratio of lavageable API to HNE at the time of HNE instillation was 4 . 8 as compared with 0 . 78 for 50 % inhibition of elastolytic activity in vitro , indicating that API is only 16 % as efficient in vivo as compared with its in vitro HNE inhibitory effectiveness . ^^^ Nevertheless , we conclude that human API given intratracheally is efficacious against HNE induced emphysema and secretory cell metaplasia . . ^^^

Effects of atriopeptin 2 ( AP 2 ) at 10 ( 7 ) M on the 45Ca flux and contractile responses to vasoconstrictors including norepinephrine ( NE ) , angiotensin 2 ( angio 2 ) and high K were studied in isolated rabbit aortic strips . ^^^ Augmentation of 45Ca efflux from aorta in normal physiological saline ( PSS ) due to NE and angio 2 each at 3 10 10 ( 7 ) M was greatly inhibited by AP 2 , suggesting that stimulated increase in cytosolic Ca2+ was suppressed . ^^^ Furthermore , the marked inhibition of contractile responses to NE ( 58 % ) and angio 2 ( 57 % ) by AP 2 was accompanied by significant decreases in 45Ca influx , whereas AP 2 exhibited only a modest inhibition on KCl ( 40 mM ) induced contraction ( 28 % ) without affecting the accompanying increase in 45Ca influx . ^^^ The 45Ca efflux from aortae in Ca2+ free PSS due to NE was markedly diminished by AP 2 , suggesting impairment of intracellular Ca2+ release . ^^^ It is concluded that AP 2 is preferentially antagonistic against vascular responses to NE and angio 2 vs . high K and that inhibition of Ca entry and release forms the primary basis of its potent vasorelaxant action against vasoconstrictors . . ^^^
