Like EGF URO , TGF alpha was a potent stimulator of femoral arterial blood flow in vivo ; and , when added to the tissue bath in vitro before an agonist , TGF alpha like EGF URO was a potent inhibitor of the contractile responses of helical coronary arterial strips to various smooth muscle agonists : norepinephrine ( NE ) , prostaglandin F 2 alpha ( PGF alpha ) and potassium chloride ( KCl ) . ^^^ The inhibitory action of EGF URO for agonist ( NE , PGF 2 alpha or KCl ) mediated contraction was diminished with repeated exposure of the tissue to EGF URO , whereas the inhibitory action of TGF alpha persisted with repetitive tissue exposure . Activated neutrophils recruited to the airways ( and their secreted products ) play several key roles in EGFR dependent mucus hypersecretion : ( 1 ) activated neutrophils secrete tumor necrosis factor ( TNF ) alpha , which induces EGFR expression in airway epithelial cells ; ( 2 ) activated neutrophils release reactive oxygen species , which activate EGFR ; ( 3 ) neutrophil elastase cleaves the EGFR proligand , pro transforming growth factor ( TGF ) alpha , releasing mature TGF alpha which activates EGFR in a ligand dependent fashion ; and ( 4 ) neutrophil elastase causes potent goblet cell degranulation . ^^^

RESULTS : We demonstrate that in BLM treated mice ( 1 ) the development of elastolytic emphysema precedes that of fibrosis ; ( 2 ) significant amount of elastase in alveolar interstitium is associated with an increased expression of TGF beta and TGF alpha ; and finally , ( 3 ) emphysematous and fibrotic lesions can be significantly attenuated by using a protease inhibitor active against neutrophil elastase . ^^^

Previous studies showed that HNE induces MUC5AC mucin production involving reactive oxygen species ( ROS ) generation and TGF alpha dependent epidermal growth factor receptor ( EGFR ) activation in human airway epithelial cells . ^^^ We hypothesize that HNE activates TACE via ROS generation , resulting in cleavage of pro TGF alpha , EGFR activation , and MUC5AC mucin expression in airway epithelial cells . ^^^ Here we show that in human airway epithelial cells HNE increases TGF alpha release , EGFR phosphorylation , and MUC5AC mucin expression , effects that were attenuated by TACE inhibitor TAPI 1 and by specific knockdown of TACE expression with small interfering RNA , implicating TACE in HNE induced responses . ^^^
