Using constructs ( COL1A2 / CAT ) containing the promoter for the alpha 2 ( 1 ) collagen gene in transient transfection assays with matched pairs of scleroderma and normal skin fibroblasts , we observed higher transcriptional activity of the COL1A2 gene in scleroderma fibroblasts and , in contrast to normal fibroblasts , no further expression was observed in the presence of TGF beta 1 . ^^^

We have previously identified a strong TGF beta 1 responsive element ( TbRE ) in the upstream promoter sequence of the alpha 2 ( 1 ) collagen ( COL1A2 ) gene . ^^^ Our experiments have shown that TGF beta 1 stimulates COL1A2 transcription by increasing binding of an Sp 1 containing complex ( TbRC ) to the TbRE . ^^^ Here , we provide evidence showing that tyrosine dephosphorylation of nuclear proteins mimics the stimulation of COL1A2 transcription by the TGF beta 1 activated signaling pathway . ^^^

BACKGROUND : Transforming growth factor beta 1 ( TGF beta 1 ) induces alpha 2 ( 1 ) collagen gene ( COL1A2 ) expression in mesangial cells through physical and functional cooperation of Smad proteins and Sp 1 . ^^^ RESULTS : TGF beta 1 induced COL1A2 gene expression in cultured mouse mesangial cells which was blocked by overexpression of inhibitory Smad 7 . ^^^ TGF beta 1 induced interaction of endogenous p 300 with Smad2 / 3 , and a dominant negative construct of p 300 inhibited the TGF beta 1 induced COL1A2 expression in cultured mouse mesangial cells . ^^^
