Mutations in XPA that prevent association with ERCC 1 are defective in nucleotide excision repair . ^^^ The human repair proteins XPA and ERCC 1 have been shown to be absolutely required for the incision step of nucleotide excision repair , and recently we identified an interaction between these two proteins both in vivo and in vitro ( L . ^^^ The ERCC 1 binding domain on XPA was previously mapped to a region containing two highly conserved XPA sequences , Gly 72 to Phe 75 and Glu 78 to Glu 84 , which are termed the G and E motifs , respectively . ^^^ In vitro , the binding of ERCC 1 to delta E was reduced by approximately 70 % , and binding to delta G was undetectable ; furthermore , both mutants failed to complement XPA cell extracts in an in vitro DNA repair synthesis assay . ^^^ These results suggest that the association between XPA and ERCC 1 is a required step in the nucleotide excision repair pathway and that the probable role of the interaction is to recruit the ERCC 1 incision complex to the damage site . ^^^

Messenger RNA levels of XPAC and ERCC 1 in ovarian cancer tissue correlate with response to platinum based chemotherapy . ^^^ The rate limiting step , DNA damage recognition and excision , is effected by the protein products of the genes ERCC 1 and XPAC . ^^^ We therefore assessed mRNA levels of ERCC 1 and XPAC in malignant ovarian cancer tissues from 28 patients that were harvested before the administration of platinum based chemotherapy . ^^^ Cancer tissues from patients whose tumors were clinically resistant to therapy ( n = 13 ) showed greater levels of total ERCC 1 mRNA ( P = 0 . 059 ) , full length transcript of ERCC 1 mRNA ( P = 0 . 026 ) , and XPAC mRNA ( P = 0 . 011 ) , as compared with tumor tissues from those individuals clinically sensitive to therapy ( n = 15 ) . ^^^ These data suggest greater activity of the DNA excision repair genes ERCC 1 and XPAC in ovarian cancer tissues of patients clinically resistant to platinum compounds . ^^^

Cell cycle checkpoint abrogator UCN 01 inhibits DNA repair : association with attenuation of the interaction of XPA and ERCC 1 nucleotide excision repair proteins . ^^^ Because mammalian cells remove CDDP induced DNA adducts through the nucleotide excision repair ( NER ) pathway , we determined the effects of UCN 01 on NER by measuring its effects on the interaction of the repair factors XPA and ERCC 1 and the phosphorylation / dephosphorylation of the repair proteins . ^^^ Although expression of XPA and ERCC 1 proteins was elevated in cells exposed to UCN 01 , the treatment resulted in a decreased ERCC 1 level in the Triton 10 100 insoluble fraction of cell lysates . ^^^ A pull down assay using the MBP XPA fusion protein showed a significant reduction in the binding of ERCC 1 to XPA in nuclear extracts from UCN 01 treated cells compared with untreated cells , suggesting that UCN 01 reduced the XPA ERCC 1 interaction . ^^^ In vitro phosphorylation revealed that UCN 01 had no effect on the phosphorylation / dephosphorylation status of either XPA or ERCC 1 ; however , UCN 01 caused dephosphorylation of an unidentified XPA bound protein with an apparent molecular mass of 52 kDa . ^^^

We have now determined genotypes for seven single nucleotide polymorphisms in five nucleotide excision repair genes , including Xeroderma pigmentosum complementation group A ( XPA +62T > C ) , group C ( XPC Lys939Gln and Ala499Val ) , group D ( XPD Asp312Asn and Lys751Gln ) , and group G ( XPG His1104Asp ) and ERCC 1 ( 8092 C > A ) in a total of 160 sister pairs for whom DRC phenotype data were available . ^^^
