The importance of regulated proteolysis in epithelia is well demonstrated by the recent identification of the SPINK 5 serine proteinase inhibitor as the defective gene in Netherton syndrome , cathepsin C mutations in Papillon Lefevre syndrome , cathepsin L deficiency infurless mice , targeted ablation of the serine protease Matriptase / MTSP1 , targeted ablation of the aspartate protease cathepsin D , and the phenotype of targeted epidermal overexpression of stratum corneum chymotryptic enzyme in mice . ^^^

The results suggest that cathepsin L could be an important activator of DPPI in vivo and that cathepsin D and possibly the cystatins may contribute to DPPI downregulation . . ^^^

To ascertain the critical thresholds of intracranial pressure ( ICP ) and cerebral perfusion pressure ( CPP ) for cerebral circulation and brain function , the extra and intracranial haemodynamics and electrical brain responses were evaluated noninvasively with Doppler ultrasonography and multimodality evoked potentials ( MEP ' s ) in 50 patients with severe head injury . ^^^ The frequency and degree of abnormalities of MEP ' s were proportionally increased with the rise of ICP and reduction of CPP . ^^^

Continuing our theoretical studies of the oxytocin and vasopressin analogues , we have analysed the molecular electrostatic potential ( MEP ) and the norm of the molecular electrostatic field ( MEF ) of [ 1 beta mercaptopropionic acid ] arginine vasopressin ( [ Mpa 1 ] AVP ) , [ 1 ( beta mercapto beta , beta cyclopentamethylene ) propionic acid ] arginine vasopressin ( [ Cpp ' ] AVP ) , and [ 1 thiosalicylic acid ] arginine vasopressin ( [ Ths ' ] AVP ) whose low energy conformations were calculated in our previous work . ^^^ The results obtained show that the shapes of both MEP and MEF are similar in the case of [ Mpa 1 ] AVP and [ Cpp 1 ] AVP ( biologically active ) , while some differences emerge when comparing these compounds with [ Ths 1 ] AVP ( inactive ) . ^^^

Four phthalate esters , namely , di ( 2 ethylhexyl ) phthalate ( DEHP , di n pentyl phthalate ( DPP ) . , di n octyl phthalate ( DOP ) , and diethyl phthalate ( DEP ) were investigated in vivo and their monoesters ( MEHP , MPP , MOP , and MEP , respectively ) in vitro for indications of Leydig cell toxicity in the rat . ^^^

To test this possibility , we examined the activities of aminopeptidase M ( APM ) , dipeptidyl peptidase 4 ( DPP 4 ) , metalloendopeptidase 24 . 15 ( MEP 24 . 15 ) and neutral endopeptidase 24 . 11 ( NEP 24 . 11 ) in rat brain , heart , kidney , liver , lung and thoracic aorta . ^^^ In contrast , in aorta , the activity of APM was slightly but significantly reduced whereas those of DPP 4 and MEP 24 . 15 were markedly enhanced ; NEP 24 . 11 was not detected in this tissue . ^^^ Immunoblotting for DPP 4 and MEP 24 . 15 showed increased expression in aortic tissue . ^^^ Neither L NAME ( 1 100microM ) nor the NO donors sodium nitroprusside and 3 morpholinosydnonimine ( SIN 1 ; 1 100microM ) had any consistent effect on the activity of recombinant MEP 24 . 15 or renal DPP 4 . ^^^

Cathepsin J represents the murine homologue of a previously described rat cathepsin L related protein . ^^^ Mature cathepsin J shows 59 . 3 % identity to mouse cathepsin L and contains the characteristic ER ( F / W ) NIN motif within the propeptide indicating that this protease belongs to the subgroup of cathepsin L like cysteine proteases . ^^^ Ctsj was mapped to mouse chromosome 13 in the vicinity of cathepsin L suggesting that cathepsin J may have arisen by gene duplication from cathepsin L or a common ancestral gene . . ^^^
