| A cdc 15 like adaptor protein ( CD2BP1 ) interacts with the CD 2 cytoplasmic domain and regulates CD 2 triggered adhesion . ^^^ A human CD 2 cytoplasmic tail binding protein , termed CD2BP1 , was identified by an interaction trap cloning method . ^^^ Although the CD2BP1 SH 3 domain binds directly to the CD 2 sequence , KGPPLPRPRV ( amino acids 300 309 ) , its association is augmented markedly by the CD2BP1 N terminal segment . ^^^ Upon ligand induced clustering of surface CD 2 molecules , CD2BP1 redistributes from a cytosolic to a surface membrane compartment , co localizing with CD 2 . ^^^ In turn , CD 2 stimulated adhesion is downregulated by CD2BP1 , apparently through coupling of the protein tyrosine phosphatase ( PTP ) PEST to CD 2 . ^^^ |

| Prominent in T cells and natural killer cells , CD 2 binding protein 1 ( CD2BP1 ) plays an important role in CD 2 mediated adhesion and signal transduction . ^^^ In the current study , we investigated CD 2 and PSTPIP ( proline , serine , threonine phosphatase interacting protein , murine homologue of CD2BP1 ) interactions in purified mouse splenic T cells . ^^^ Regulation of the association between PSTPIP and CD 2 in murine T cells . ^^^ PSTPIP associated with CD 2 in both resting and activated T cells . ^^^ Following various stimuli , such as concanavalin A , anti TCRbeta , anti CD3epsilon , anti CD3epsilon / phorbol myristate acetate ( PMA ) , IL 2 , or PMA / ionomycin , PSTPIP and CD 2 expression , as well as their association , increased in a time dependent fashion . ^^^ |

| The Wiskott Aldrich syndrome protein acts downstream of CD 2 and the CD2AP and PSTPIP 1 adaptors to promote formation of the immunological synapse . ^^^ Here , we show that antigen induced formation of T cell : APC conjugates and synapses is abrogated in WASp deficient T cells and that CD 2 engagement evokes interactions between the proline rich region required for WASp translocation to the synapse and the PSTPIP 1 adaptor SH 3 domain and between the PSTPIp 1 coiled coil domain and both CD 2 and another CD 2 binding adaptor , CD2AP . ^^^ These data , together with the impairment in CD 2 induced actin polymerization observed in WASp deficient cells , suggest that PSTPIP 1 acts downstream of CD2 / CD2AP to link CD 2 engagement to the WASp evoked actin polymerization required for synapse formation and T cell activation . . ^^^ |

| CD2BP1 modulates CD 2 dependent T cell activation via linkage to protein tyrosine phosphatase ( PTP ) PEST . ^^^ This study focuses on CD2BP1 , a CD 2 cytoplasmic tail binding protein preferentially expressed in hematopoetic cells . ^^^ In this study , using a murine Ag specific primary T cell transduction system to assess CD 69 , IL 2 , and IFN gamma expression , we provide evidence that CD2BP1 directly and negatively impacts T cell activation via isolated CD 2 triggering or TCR stimulation dependent on coordinate CD 2 engagement . ^^^ Disruption of protein tyrosine phosphatase PEST and / or CD2BP1 association with the CD 2 signalsome rescues T cells from the inhibitory effect of CD 2 crosslinking . ^^^ The overexpression of CD2BP1 selectively attenuates phospholipase Cgamma 1 , ERK1 / 2 , and p 38 phosphorylation without abrogating CD 2 independent TCR stimulation . ^^^ |
