As recently S 100 proteins have been identified as ligands of RAGE , we sought to determine the effects of the proinflammatory heterodimer of S100A8 / S100A9 on the RAGE NFkappaB mediated induction of proinflammatory gene expression . ^^^ Following this preactivation , cells were stimulated for 48 h with heterodimeric human recombinant S100A8 / S100A9 . ^^^ RESULTS : Heterodimeric S100A8 / S100A9 enhanced secretion of IL 6 , ICAM 1 , VCAM 1 and MCP 1 in AGE albumin pretreated HUVEC in a dose dependent manner . ^^^ These effects could not be detected after stimulation with the homodimeric proteins S100A8 , S100A9 , S100A1 and S100B . ^^^ The effects of heterodimeric S100A8 / S100A9 were reduced by inhibition of the MAP kinase pathways ERK1 / 2 and p 38 by PD 98059 and SB 203580 , respectively . ^^^

S100A8 and S100A9 , two Ca2+ binding proteins of the S 100 family , are secreted as a heterodimeric complex ( S100A8 / A9 ) from neutrophils and monocytes / macrophages . ^^^ Serum and synovial fluid levels of S100A8 , S100A9 , and S100A8 / A9 were all higher in patients with rheumatoid arthritis ( RA ) than in patients with osteoarthritis ( OA ) , with the S100A8 / A9 heterodimer being prevalent . ^^^ S100A8 / A9 complexes , as well as S100A9 homodimers , stimulated the production of proinflammatory cytokines , such as tumor necrosis factor alpha , by purified monocytes and in vitro differentiated macrophages . ^^^

METHODS : The lungs of 20 day old congenic C57BL / 6 ( before any overt signs of inflammation ) and BALB / c CF mouse lines maintained in sterile environments were investigated for distinctions in the neutrophil chemokines S100A8 and S100A9 by quantitative RT PCR and RNA in situ hybridization , that were then correlated to neutrophil numbers . ^^^ RESULTS : The lungs of C57BL / 6 CF mice had spontaneous and significant elevation of both neutrophil chemokines S100A8 and S100A9 and a corresponding increase in neutrophils , in the absence of detectable pathogens . ^^^ In contrast , BALB / c CF mouse lungs maintained under identical conditions , had similar elevations of S100A9 expression and resident neutrophil numbers , but diverged in having normal levels of S100A8 . ^^^ CONCLUSION : The results indicate early and spontaneous lung inflammation in CF mice , whose progression corresponds to increased expression of both S100A8 and S100A9 , but not S100A9 alone . ^^^ Moreover , since both C57BL / 6 and BALB / c CF lungs were maintained under identical conditions and had similar elevations in S100A9 and neutrophils , the higher S100A8 expression in the former ( or suppression in latter ) is a result of secondary genetic influences rather than environment or differential infection . . ^^^

BACKGROUND AND OBJECTIVE : Calprotectin is composed of two proteins , S100A8 and S100A9 , which are S 100 family members , and is detected in gingival crevicular fluid and gingival tissue with inflammation . ^^^ The expression of S100A8 / S100A9 mRNAs was examined by northern blotting and the amount of calprotectin was measured by enzyme linked immunosorbent assay ( ELISA ) . ^^^ RESULTS : Norepinephrine stimulated the expression of S100A8 / S100A9 mRNAs via beta adrenergic receptors in U 937 cells and significantly increased calprotectin production to about 3 . 6 fold that of the control . ^^^

Calcium dependent tetramer formation of S100A8 and S100A9 is essential for biological activity . ^^^ The heterodimers of S100A8 and S100A9 represent the major calcium binding proteins in phagocytes . ^^^ Calcium binding induces formation of ( S100A8 / S100A9 ) 2 tetramers . ^^^ Mutations in EF hand 2 did not interfere with formation of the S100A8 / S100A9 heterodimer as demonstrated by yeast two hybrid experiments and pull down assays . ^^^ In contrast , mass spectrometric analysis and density gradient centrifugation revealed that calcium induced association of ( S100A8 / S100A9 ) 2 tetramers was strictly dependent on a functional EF hand 2 in S100A9 . ^^^

OBJECTIVES : We studied whether serum levels of S 100 proteins A 8 ( S100A8 ) and A 9 ( S100A9 ) are elevated in psoriasis , correlated their amounts with disease activity and identified potential cellular sources . ^^^ METHODS : Serum obtained from psoriasis patients or from healthy individuals was studied for S100A8 and S100A9 levels by enzyme linked immunosorbent assay . ^^^ Cellular sources of S100A8 and S100A9 were identified by in situ hybridization and immunohistochemistry of lesional psoriatic and nonlesional , nonpsoriatic skin . ^^^ RESULTS : A significant increase of S100A8 / S100A9 serum levels was found in patients with psoriasis compared with healthy controls . ^^^ Performing in situ hybridization of lesional psoriatic skin we detected a dramatic induction of both S100A8 and S100A9 mRNA and protein primarily in the suprabasal layers of the epidermis while expression was negligible in nonlesional , nonpsoriatic interfollicular epidermis . ^^^
