Cyclin D 1 ( CCND 1 ) and retinoblastoma ( Rb ) genes are cell cycle regulators which are altered in some breast carcinomas . ^^^ However , the possible cooperation between CCND 1 and Rb , as well as the influence and coincidence of their abnormalities in the proliferative capacity of mammary carcinoma cells in vivo , is still unknown . ^^^ In order to assess both the significance of the CCND 1 gene and Rb alterations in breast carcinomas and their relationship with the proliferative capacity of the tumours and other clinico pathological factors , CCND 1 mRNA expression was studied in 46 cases of primary breast carcinomas and matched normal tissue , 45 of which were also studied immunohistochemically , Rb expression was analysed in the same cases by immunohistochemistry , whereas the proliferative activity of the carcinoma was evaluated by flow cytometry . ^^^ This abnormality was significantly associated with increased mean S phase ( P = 0 . 017 ) and decreased CCND 1 mRNA expression in non overexpressed tumours , supporting in vivo the postulated regulatory loop between Rb and CCND 1 in vitro . ^^^ We conclude that CCND 1 up regulation is not associated with increased proliferative activity in breast carcinomas , whereas its expression might be regulated in vivo by hormones and Rb . ^^^

To investigate whether alteration of other components in pRb pathway is an alternative mechanism underlying the development of these chemically induced lymphomas , we have examined the genetic status of Rb 1 , Ccnd 1 and Cdk 4 genes that encode retinoblastoma protein , cyclin D 1 and cyclin dependence kinase 4 , respectively . ^^^ Gross alterations of the Rb 1 , Ccnd 1 , and Cdk 4 genes were not detected by Southern analysis in any of the tumors examined . ^^^ These results indicate that the mechanisms underlying the development of 2 ' , 3 ' dideoxycytidine and 1 , 3 butadiene induced lymphomas involve inactivation of p16 / p15 cyclin dependent kinase inhibitors but not genomic alterations of the Rb 1 , Ccnd 1 and Cdk 4 genes . . ^^^

Differential alterations of the genes in the CDKN2A CCND 1 CDK4 RB 1 pathway are associated with the development of head and neck squamous cell carcinoma in Indian patients . ^^^ RESULTS : The alterations [ mutation , hypermethylation , homozygous deletion and loss of heterozygosity / microsatellite size alteration ( LOH / MA ) ] in the CDKN2A were found to be highest in 57 % of the samples , followed by CCND 1 amplification and LOH / MA at the RB 1 locus in 14 % and 8 . 5 % of the samples , respectively . ^^^ Comparatively high frequency of CDKN2A alterations ( except homozygous deletion ) was found in dysplastic head and neck lesions and remained almost constant or increased during progression of the tumour , whereas the homozygous deletion of CDKN2A and the alterations in CCND 1 and RB 1 genes were seen mainly in the later stages of the tumour . ^^^
