An improved three step Percoll density gradient centrifugation technique is described for simultaneous isolation of glial plasmalemmal vesicles ( GPV ) and synaptosomal vesicles ( SYN ) from a rat brain homogenate . ^^^ While electron microscopy revealed that fractions contained intact vesicles with markedly distinct morphological features , measures of high affinity [ 3H ] choline uptake , glutamine synthetase and carbonic anhydrase activities , as well as Western blot analyses for glial fibrillary acidic protein and neuron specific enolase , served to confirm the low level of neuronal contamination in GPV fractions as well as the low level of glial contamination in SYN fractions . ^^^ In addition , GPV and SYN fractions were used to characterize the kinetic and pharmacological properties of sodium dependent [ 3H ] L glutamate transport . ^^^ |

Using coprecipitation methods with convulxin coupled beads and antibodies against FcRgamma and the Src family , we showed that Fyn and Lyn , but not Yes , Src , Fgr , Hck , and Lck , were physically associated with the GPVI FcRgamma complex irrespective of stimulation . ^^^ Furthermore , Fyn was rapidly activated by collagen or cross linking GPVI . ^^^ |

The glycoprotein 6 ( GPVI ) Fc receptor ( FcR ) gamma chain complex , a key activatory receptor for collagen on platelet surface membranes , is constitutively associated with the Src family kinases Fyn and Lyn . ^^^ Molecular cloning of GPVI has revealed the presence of a proline rich domain in the sequence of GPVI cytoplasmic tail which has the consensus for interaction with the Src homology 3 ( SH 3 ) domains of Fyn and Lyn . ^^^ Furthermore , depletion of the proline rich domain in GPVI ( Pro ( ) GPVI ) prevented binding of Fyn and Lyn and markedly reduced phosphorylation of FcR gamma chain in transiently transfected COS 7 cells , but did not affect the association of the gamma chain with GPVI . ^^^ These findings demonstrate that the proline rich domain of GPVI mediates the association with Fyn / Lyn via their SH 3 domain and that this interaction initiates activation signals through GPVI . . ^^^ |
