| Association of the Src family tyrosine kinase Fyn with TrkB . ^^^ The Fyn Src homology domain 2 ( SH 2 ) was observed to associate in vitro with the intracellular domain of TrkB ( ICD TrkB ) . ^^^ The Fyn SH 2 domains bound to phosphorylated ICD TrkB ( pICD TrkB ) with an affinity similar to the binding of phospholipase Cgamma ( PLCgamma ) SH 2 domains to its autophosphorylation site in TrkB . ^^^ The Src SH 2 domains showed substantially lower affinity with pICD TrkB , suggesting that the association between Fyn SH 2 and pICD TrkB is not due to nonspecific interactions of SH 2 domains with phosphorylated tyrosine residues . ^^^ This is further supported by the observation that Fyn SH 2 was able to trap phosphorylated TrkB in cell lysate prepared from primary rat cortical neurons stimulated with brain derived neurotrophic factor ( BDNF ) . ^^^ |

| Here , we show that Fyn is a key molecule linking the BDNF receptor TrkB with NMDA receptors , which play an important role in spatial memory formation in a radial arm maze . ^^^ Spatial learning induced phosphorylation of TrkB , Fyn , and NR2B , but not NR2A , in the hippocampus . ^^^ Fyn was coimmunoprecipitated with TrkB and NR2B , and this association was increased in well trained rats compared with control animals . ^^^ The phosphorylation of Fyn and NR2B , but not TrkB , was diminished by PP 2 treatment . ^^^ |
