Here we performed experiments to identify which cellular protein phosphatase ( PP 1 , PP2A , or PP2B ) is responsible for the in vivo phosphorylation / dephosphorylation of Hsp 27 . ^^^ In their purified forms , PP2A dephosphorylates Hsp 27 more effectively than PP2B , whereas PP 1 is weakly active . ^^^ However , the phosphorylation of Hsp 27 in vivo is only affected when cells are treated with PP 1 and PP2A inhibitors ( okadaic acid , calyculin A ) or cantharidin ( PP2A inhibitor ) , but not the PP2B inhibitor , cyclosporin A , suggesting PP2A to be the main enzyme dephosphorylating Hsp 27 in the cells . ^^^ Purification and immunoblotting of Hsp 27 phosphatase from MRC 5 cells also suggest it to be PP2A and not PP 1 or PP2B . ^^^

Protein phosphatase ( PP ) inhibitors ( okadaic acid , cantharidin , sodium orthovanadate ) did not delay the ATP decrease in energy deprived EC but arrested both the alterations in the Hsp 27 status and the changes for the worse in F actin and cell morphology . ^^^ The longer preservation of the cytosolic pool of phosphorylated Hsp 27 during ATP depletion in the PP inhibitor treated or thermotolerant EC correlated with the acquired resistance of F actin and morphology . ^^^

In addition , immunoblot analysis demonstrated that detection of total Hsp 27 increased ( P < 0 . 05 ) between day 21 and 1 day post partum ( PP ) inclusive . ^^^ Immunocytochemical analysis of Ser 15 phosphorylated Hsp 27 expression in situ demonstrated that phosphorylated Hsp 27 in circular muscle became detectable in peri nuclear and membrane regions on days 19 to 22 , but was primarily restricted to the cytoplasm on days 23 to PP . ^^^ In contrast , phosphorylated Hsp 27 in longitudinal muscle was primarily detected in myocyte membranes on days 15 to 22 , and then also became detectable in the cytoplasm of myocytes on days 23 and PP . ^^^
