Dissociation of eIF4F was enhanced during heat shock by ectopic overexpression of Hsp 25 , the murine homolog of human Hsp 27 . ^^^ Hsp 27 specifically bound eIF4G during heat shock , preventing assembly of the cap initiation / eIF4F complex and trapping eIF4G in insoluble heat shock granules . eIF4G is a specific target of Hsp 27 , as eIF4E , eIF4A , Mnk 1 , poly ( A ) binding protein , eIF4B , and eIF 3 were not bound by Hsp 27 and were not recruited into insoluble complexes . ^^^ These results therefore demonstrate that Hsp 27 is a heat induced inhibitor of eIF4F dependent mRNA translation . . ^^^ |

The proteasome inhibitor , MG 132 , promotes the reprogramming of translation in C2C12 myoblasts and facilitates the association of hsp 25 with the eIF4F complex . ^^^ Prolonged incubation with MG 132 resulted in the increased expression of heat shock protein ( hsp ) 25 , alphaB crystallin and hsp 70 , with a population of hsp 25 associating with the eIF4F complex in a p 38 mitogen activated protein kinase dependent manner . ^^^ Addition of recombinant hsp 25 to an in vitro translation assay resulted in stimulation of on going translation and a moderate decrease in de novo translation , indicating that this modified eIF4F complex containing hsp 25 has a role to play in recovery of mRNA translation following cellular stress . . ^^^ |

Hsp 27 also partially prevented the sharp decrease in the pool of mobile cytoplasmic EGFP eIF4G . sHsps did not prevent the phosphorylation of eIF2alpha by a heat shock , but promoted dephosphorylation during recovery . ^^^ Our data show that sHsps do not prevent the inhibition of protein synthesis upon heat shock , but restore translation more rapidly by promoting the dephosphorylation of eIF2alpha and , in the case of Hsp 27 , the availability of eIF4E and eIF4G . . ^^^ |
